In this study, it was found that Rubescensine A undergoes a classical nucleophilic addition reaction with Cys145 of 3CLpro, and the sulfhydryl group of Cys145 acts as a nucleophilic group to chemically react with α, β-unsaturated olefins on Rubescensine A to form The addition product of it-Cys145 blocks the binding of 3CLpro to the substrate, thereby achieving the purpose of inhibiting the virus.

The study found that Oridonin A can exert an anti-SARS-CoV-2 effect by targeting the inhibition of 3CLpro. Interestingly, the 3CLpro sequences of several different major new coronavirus mutant strains (including the current epidemic strains) were found to be very conservative through gene sequence comparison, and the only two mutation sites that appeared so far were not bound by Rubescensine A. locus, suggesting that Rubescensine A should also have a strong inhibitory effect on the currently circulating Omicron strains.
